Hmmm, based on this document: http://support.f5.com/kb/en-us/products/em/manuals/product/em-getting-started-3-1-1/ only LDAP, Active Directory, RADIUS or TACACS+ are supported it seems.
SecurID Authentication Manager can be configured to run a RADIUS server, so you should be able to use that to protect Enterprise Manager with RSA two-factor.
RSA is inherently two-factor. It's something you know (the PIN) and something you have (the tokencode).
