That's a bit light on the information there...
Which monitor? Does the monitor have debugging available? If so, switch the debug on. What do you see?
If you tcpdump the interface between the BigIP and the pool member, do you see the monitor traffic? What's in it?
When you say the pool member is down, how do you mean? The server is shutdown completely? The IP isn't on the network? The web server (Or whatever) isn't running but the host is up?
You'll need to provide a fair bit more info..

Health monitor which we assign to the pool member's.. Like dat we have configured monitor on port 2500 and assign to the pool member..
We are not getting any response from backends on port 2500..

Anuj, please put some answers to below, so that I can narrow down the issue-
1) Are you able to telnet to pool member from LTM on port 2500?
2) What do you see in packet capture/tcpdump?
3) Is it a custom monitor? Is there any Aias IP/POrt defined into that monitor?
4) Is it happening on all the monitors you configured? Or specifically for port 2500's monitor?
5) Can you paste the configuration out put of monitor and pool here (tmsh list ltm pool AND tmtsh list ltm monitor )

Yes able to telnet..
Tcpdump.shows monitor does not work.
Even f5 tac also confirm the same.
For all moniotre we configured basically which are not firing..
